Blood is made up of blood cells and plasma, and blood cells are made up of red blood cells, white blood cells and platelets, no anticoagulants are added, centrifugation separates light yellow liquid which is called serum.
Serum contains active substances such as proteins, enzymes, amino acids, vitamins, etc., which are mainly used for immune assessment and disease diagnosis.
Whole blood samples are needed to be pre-treated and separating serum by using a low-speed centrifuge. This is currently the most efficient method. So how to isolate serum correctly?

Experimental Procedure
Collect blood samples, put them into the centrifuge tube, gently mix them 5 or 6 times.No violent shaking here in order to avoid hemolysis, and leave it at room temperature for about 15 minutes, the blood in the tube will be coagulated and does not flow.
Place the centrifuge tube containing the whole blood sample into the centrifuge and set speed to 3000-3500 rpm for 5-10 min.
After centrifugation, use a micro pipette and pipette tips to slowly transfer the serum to a sterile centrifuge tube.
Storage of serum requires a further temperature of -20°C or lower, which is able to maintain stability and antigenic activity.
Precautions
●The whole process of operation shall be under sterile environment to avoid contamination of experimental samples
●The speed and time of the centrifuge need to be determined according to samples
●After collecting the blood, separate serum and plasma as soon as possible to avoid hemolysis, and repeated freezing and thawing of serum and plasma.
●Blood centrifugation speed should not be too fast, as too fast centrifugation can cause hematocrit and hemolysis.
●If the blood sample cannot be separated from serum or plasma immediately, it should be stored and transported as refrigerated as possible, and should never be frozen. Otherwise, the blood cells will form ice crystals, causing the cells to rupture and cause hemolysis.
